Two things work against you here.
First, the buildings. Venetian hotels live inside Gothic and Renaissance palazzi with protected floor plans, load-bearing walls you can't move, and no room to add an extension. When a Venice hotel wants to sleep four, it almost always does one of three things: a king plus a sofa bed, a king plus two rollaway singles, or a two-bedroom suite. A room with two full-size beds side by side is the exception, not the rule.
Second, the culture. Italian hotels are built around the letto matrimoniale, one double bed for a couple, and letti singoli for children. The idea of two adult-sized beds in a single room for a family of four isn't how Italian hoteliers think.
Here's the good news, and it's the part most American families don't know: an Italian "double" bed is bigger than a US queen. A standard matrimoniale is 160 cm wide, about 63 inches, versus 60 inches for a US queen. So when a Venice hotel lists a room with "two double beds," you're getting two beds each slightly wider than a queen at home. (The narrower 140 cm letto alla francese, about 55 inches, is closer to a US full. I've noted below where that matters.)

The Venice Times Hotel, Vignette Collection by IHG, Cannaregio
Look for the Superior Queen Room with two queen beds. This is the one property on this list where I can point to a guest review confirming exactly what you're asking for: a family of two adults and two small children fitting comfortably into two queen beds in one room, with staff described as fantastic with kids. The hotel sits down a quiet alley a block off the Grand Canal, next door to the main train station and the bus line into the city, which makes it one of the easiest Venice hotels to reach with luggage and children in tow. It's an intimate property, around 30 rooms, styled with Venetian damask and velvet rather than grand palazzo scale, and the rate reflects that: this is the value pick on the list, not the splurge. Hotel Metropole, Riva degli Schiavoni (Castello)
Look for the Damasco Exclusive Suite, Lagoon View, which is configured with two double (Italian matrimoniale, wider than a US queen) beds. The Metropole is one of my favorite hotels in Venice for reasons that have nothing to do with beds: it occupies the building where Vivaldi taught music to orphan girls at the Ospedale della Pietà, and the owning Beggiato family has filled it with their collections of antique crucifixes, fans, corkscrews and nutcrackers, so the corridors feel like a cabinet of curiosities. It's a five-minute walk along the waterfront from Piazza San Marco, with the San Zaccaria vaporetto stop out front and direct Alilaguna airport boats. For larger families, the hotel's annex San Giorgio Apartment sleeps five with two doubles and a single, and gives you a kitchen. The Oriental Bar does one of the best afternoon teas in the city.

Sina Centurion Palace, Dorsoduro
Look for the Presidential Suite, Grand Canal View, which is the only room I've found in a Venice 5-star with two king beds in it (sleeps 5). This is the top of the market: a 1,292-square-foot suite in the Palazzo Genovese, right next to the Salute church with the Grand Canal at your feet and the Guggenheim around the corner. The hotel is small (50 rooms), contemporary inside a Gothic shell, and has a private water entrance so your taxi from the airport pulls straight up to the door. If two queens is the requirement and budget is not the constraint, this is the answer. It's also a Small Luxury Hotels of the World property, so I can add value to the stay.

Family Resort Alternative: JW Marriott Venice Resort & Spa
I need to be honest here: the JW does not have a standard room with two queen beds. What it has is the Family Premium King and Family Premium Twin rooms (484 sq ft, sleep 4), more than double the size of a typical Venice hotel room, plus the two-bedroom Villa Rose with two double beds. It's on its own 40-acre island with three pools (one just for families), a kids' club that does Venetian mask decorating and pizza-making, and a free boat to San Marco every half hour that takes about 20 minutes. For a family with kids under 10, I'd rather put you here in a Family Premium room than squeeze you into a palazzo. Hotel Palazzo Stern, Dorsoduro (Grand Canal)
Look for the Junior Suites, which several past guests report are set up with two queens. This Venice hotel with two Queen beds in one room sits right on the Grand Canal at the Ca' Rezzonico vaporetto stop, which is the sweet spot of Venice for families: ten minutes' walk to the Accademia and the Guggenheim, far enough from San Marco that you can actually relax at night. The palazzo was rebuilt in the early 1900s by the Stern family in full neo-Gothic Venetian style, with mosaic floors and a small garden terrace directly on the water where you have breakfast watching the boats. It's a 4-star, not a 5-star, so don't expect a spa or a Michelin restaurant, but the location and that room configuration make it my first call for a family of four in Venice. Because it's an independent property I can't layer on Virtuoso amenities here, but I can still book it and handle everything around it, from water taxi transfers to the private Doge's Palace tour.

Hotel Gabrielli Venezia - Starhotels Collezione, Riva degli Schiavoni (Castello)
Look for the Family Room, Connecting Rooms category, which comes in two configurations: an 80-square-meter version that sleeps six across three double beds, and an 81-square-meter lagoon-view version that sleeps four with a double bed in one room and twins in the other. This is one of the few Venice hotels that sells a connecting-room family setup as a single bookable category rather than something you have to piece together at check-in. The Gabrielli sits on Riva degli Schiavoni just past the Metropole, in a palazzo the Perkhofer family has run for five generations, recently reopened after a full restoration with 66 rooms, marble bathrooms, and walnut furnishings. You're a few minutes from the Bridge of Sighs and the Doge's Palace, with a rooftop terrace for the adults once the kids are asleep. The honest trade-off: you're booking a connecting-room category, not a single oversized room, so it reads and prices more like two rooms than one. But for a family that wants kids in their own space rather than sharing a room, that's a feature, not a bug.

Palazzo De Polo, Cannaregio
Look for the Family Suite, a roughly 320-square-foot layout with a separate living room and bedroom, two beds, and a private bathroom. Palazzo De Polo is a small, recently renovated 4-star just off Salizada San Canzian in Cannaregio, tucked between the Church of San Canciano and Santa Maria dei Miracoli, a five-minute walk from the Rialto Bridge and the Grand Canal. It's the kind of restoration Venice does well: frescoed ceilings and coffered woodwork paired with new bathrooms and air conditioning, and guests consistently mention how quiet the rooms are for a property this central. The catch is size. This is a boutique hotel with a small number of rooms, so the Family Suite is limited inventory in a category that books up early, especially in shoulder season. I'd treat this one as a "call me the moment you have dates" property rather than a backup option.

Palazzo Veneziano, Dorsoduro (Zattere)
Look for the Duplex Family Room, a 43-square-meter, two-level layout with a canopy bed on the mezzanine and a second bed on the main floor, plus a bathroom finished in Carrara marble. Palazzo Veneziano sits on the Zattere waterfront in Dorsoduro, one vaporetto stop from San Marco, and is built and run as a genuinely child-friendly hotel rather than a historic property that happens to tolerate kids. Some of the rooms and suites have private jacuzzi tubs, which is unusual to find in Venice at this price point. The design leans modern Venetian rather than antique-filled palazzo, so if your family's mental image of Venice is frescoes and gilt mirrors, this won't match it. What you get instead is more space, a real family layout, and a location that lets you walk to the Accademia and the Guggenheim without fighting San Marco crowds.

The Venice Venice Hotel, Cannaregio (Grand Canal)
Look for a Family Loft, a 47-to-65-square-meter room with two beds split across a main floor and a mezzanine level, sleeping four. The Venice Venice occupies Palazzo Ca' da Mosto, a 13th-century palace directly on the Grand Canal near the Rialto Bridge, but the interior has nothing to do with the historic exterior. The owners, the founders of the Golden Goose fashion brand, gutted it and filled it with contemporary art and design pieces under a concept they call "Postveneziani," a deliberate break from the antique-Venice look. It's a genuine design hotel, a five-star with a cocktail bar and a courtyard, and it's every bit the Grand Canal splurge that Sina Centurion Palace is, just with a completely different aesthetic. If your family wants the art-gallery, ultra-contemporary version of a Venice stay, this is it; if you want frescoes and velvet, choose Sina Centurion Palace or the Metropole instead.

Budget-Friendly Venice Hotels with Two Queen Beds in One Room
Hilton Garden Inn Venice Mestre San Giuliano. This Hilton has the American-standard "Room, 2 Queen Beds" listed as a plain category, exactly as you'd find it in Chicago. The trade-off is location: it's on the mainland in Mestre, with the tram to Piazzale Roma in about 15 minutes. For a family that wants two real queens, a big buffet breakfast, and a Venice rate that doesn't have a comma in it, it's a very sensible base, especially the night before an early flight from Marco Polo.
Hotel Belle Epoque (Cannaregio, near the train station). A 3-star whose Quadruple Room is described by past guests as two queen beds in a split-level layout with internal stairs. It's a five-minute walk from Santa Lucia station, which makes it practical if you're arriving by train from Florence or Rome. (Note I can’t seem to find a way to book this hotel online.)
Hotel Villa Mabapa (Lido). A 4-star villa hotel on the lagoon side of the Lido whose Family Room (322 sq ft) has a double plus two singles and sleeps four, with breakfast included. Not two queens, but generous space at a fraction of central Venice prices.

Do Venice hotels have queen beds? Almost every Venice hotel room has one queen-or-larger bed; very few have two. Venetian hotels occupy historic palazzi with fixed floor plans, and Italian hotel culture is built around one double bed per couple plus single beds for children. When you find two full-size beds in one room, it's usually at a modern or American-brand property (the Hilton Garden Inn in Mestre, The Venice Times Hotel), a Lido hotel with bigger footprints (Ausonia Hungaria), or one of the handful of palazzo hotels in this post that made a deliberate decision to serve American families.
What is the difference between an Italian double bed and a US queen bed? This is the detail that changes everything. A standard Italian letto matrimoniale is 160 cm wide, about 63 inches, which is 3 inches wider than a US queen (60 inches). So a Venice room listed with "two double beds" gives you two beds bigger than queens. The exception is the letto alla francese at 140 cm (55 inches), roughly a US full, which some budget hotels use. Every property in my top picks above uses the full-size 160 cm bed or larger; where I wasn't sure, I've told you to ask.
Can a family of four share one hotel room in Venice? Yes, but you have to be specific about the room category, because the standard Venice double room is genuinely small, often under 200 square feet. Look for a room that explicitly names two beds, a connecting-room family category like the Gabrielli's, or a family suite with a second bedroom. I've listed the exact room names to ask for above. And note that some hotels cap occupancy at three adults regardless of bed count, so four adults in one room is much harder than two adults and two children.
What is the best area of Venice to stay in with a family? For most families I steer toward Dorsoduro (quiet, walkable, close to the Accademia and Guggenheim, easy vaporetto access) or the eastern end of Castello along the waterfront near the Metropole and Gabrielli, where you're five minutes from San Marco but out of the crush. I generally steer families away from staying directly around Piazza San Marco, where you'll pay the most for the least space and fight the crowds at every hour. For families with kids under 10, or for anyone visiting May through September, I'll often suggest the Lido or the JW Marriott's island instead, because a pool and a beach after a day of bridges and cobblestones changes the whole trip.
Is the Lido a good idea for families visiting Venice? It's my favorite trick for families. The Lido is a 15-minute vaporetto ride from San Marco, with real beaches, bikes, playgrounds, larger hotel rooms, and lower prices. You lose the romance of stepping out of your hotel into a medieval alley; you gain sanity.
